Protein and Amino Acid Metabolism in the Fetal- Placental Unit

چکیده

Introduction Protein synthesis and accretion are the cornerstones of growth that provide the structural framework and enzymatic machinery necessary for fetal development. The maternal uterine circulation supplies the components that the fetus uses for protein accretion. All are derived from the maternal diet or maternal tissue stores. It is important to explore the regulatory processes within the maternal, placental, and fetal compartments that determine fetal amino acid retention. In addition to tissue synthesis, use of free amino acids as oxidative fuel is extensive in the well-nourished fetus and increases significantly during periods of maternal food deprivation. The changes in fetal amino acid utilization represent a significant mechanism by which the fetus adapts to energy deprivation.

Placental uptake and transport of ACP, a neutral nonmetabolizable amino acid, in an ovine model of fetal growth restriction.

Reductions in fetal plasma concentrations of certain amino acids and reduced amino acid transport in vesicle studies suggest impaired placental amino acid transport in human fetal growth restriction (FGR). In the present study, we tested the hypothesis of an impairment in amino acid transport in the ovine model of hyperthermia-induced FGR by determining transplacental and placental retention an...

Fetoplacental transport and utilization of amino acids in IUGR--a review.

Amino acids have multiple functions in fetoplacental development. The supply of amino acids to the fetus involves active transport across and metabolism within the trophoblast. Transport occurs through various amino acid transport systems located on both the maternal and fetal facing membranes, many of which have now been documented to be present in rat, sheep and human placentas. The capacity ...

Uptake and release of amino acids in the fetal-placental unit in human pregnancies

OBJECTIVES The current concepts of human fetal-placental amino acid exchange and metabolism are mainly based on animal-, in vitro- and ex vivo models. We aimed to determine and assess the paired relationships between concentrations and arteriovenous differences of 19 amino acids on the maternal and fetal sides of the human placenta in a large study sample. METHODS This cross-sectional in vivo...

Maternal-fetal metabolism in normal pregnancy and preeclampsia.

Adaptation to pregnancy in humans involves major anatomic, physiologic and metabolic changes in the mother in order to support and provide for the nutritional and metabolic needs of the growing conceptus. Metabolically, pregnancy is marked by several important and dynamic adjustments including increased insulin resistance, hyperlipidemia and changes in protein and amino acid metabolism. In gene...

Plasmodium falciparum Malaria Elicits Inflammatory Responses that Dysregulate Placental Amino Acid Transport

Placental malaria (PM) can lead to poor neonatal outcomes, including low birthweight due to fetal growth restriction (FGR), especially when associated with local inflammation (intervillositis or IV). The pathogenesis of PM-associated FGR is largely unknown, but in idiopathic FGR, impaired transplacental amino acid transport, especially through the system A group of amino acid transporters, has ...
